Applications are invited for a temporary post of a UCD POST-DOCTORAL
RESEARCH FELLOW LEVEL 1 within UCD School of Medicine

Bechet&#**Apply on the website**;s disease (BD) is a heterogeneous
multifactorial auto-inflammatory condition characterised by recurrent
episodes of oral and genital ulceration, uveitis and skin lesions,
with less frequent involvement of the gastrointestinal tract, large
blood vessels and central nervous system. Two recent studies have
reported monogenic mucocutaneous ulcerative syndromes with
similarities to BD, in six

unrelated families caused by loss-of-function mutations in NF-kB
pathway genes.

We have identified 5 Irish families with a very similar illness,
primarily involving childhood-onset chronic oral and genital ulcers.
In the largest, a three generation family, whole exome sequencing
revealed segregation of a mutation in RELA (p**Apply on the website**)
with the condition. The mutation involves a cytosine deletion causing
a His**Apply on the website**ThrfsTer7 frameshift resulting in a
truncated protein of **Apply on the website** amino acids compared
with wild-type RELA of **Apply on the website** amino acids.
Crucially, the mutation interrupts the two C-terminal RELA
transactivating domains.

The research objectives are:
* To determine the biological effects of the RELA His**Apply on the
website**ThrfsTer7 mutation on NF-activities such as inflammation and
apoptosis.
* To use whole exome sequencing to identify potentially causative
mutations in 4 families.
* To undertake appropriate biological studies to determine
genotype-phenotype associations.

This study will give novel information on both the genetic basis and
biological mechanisms of an uncommon, but increasingly recognized
auto-inflammatory condition.

This is a research focused role, where you will conduct a specified
programme of research supported by research training and development
under the supervision and direction of a Principal Investigator. The
primary purpose of the role is to further develop your research skills
and competences, including the processes of publication in
peer-reviewed academic publications, the development of funding
proposals, the mentorship of graduate students along with the
opportunity to develop your skills in research led teaching.
